DETERMINATION OF DIAZEPAM RAT SERUM SAMPLES BY GAS CHROMATOGRAPHY

Abstract
The method is based on "High Resolution Gas Chromatographic Method for the Determination of Diazepam in Rat Serum", developed and validated at Bio-Research Laboratories. (SOP NO. BPM 253.1) The analytical method is summarized as follows: Diazepam and its internal standard were extracted from 0.1 mL rat serum with methylene chloride:pentane (v:v). The organic layer was removed and evaporated to dryness. The residue was then dissolved in ethyl acetate and injected into a gas chromatograph equipped with a 63Ni electron capture detector and a capillary column. Quantitation was by peak height ratio using a power curve for calculations.
