Nucleic acid identification technologies, like polymerase chain reaction (PCR), are being used by defence organizations for the identification of biothreat agents. Direct and immediate PCR analysis of samples suspected of containing biothreat agents may not always be possible, and thus there is a requirement to store and/or transport the sample prior to analysis. FTA® solid phase media are designed to rapidly lyse cells while trapping and protecting genetic material from nucleases, oxidation, and UV damage over long periods of time (years) when stored at room temperature. FTA® cards therefore have the potential to provide a safe, simple, and convenient method for processing, archiving, and transporting samples containing, or suspected of containing biothreat agents. A project was undertaken under the auspices of the CAN–NLD–SWE Cooperative Science and Technology MoU Project Arrangement (PA2004-04) to evaluate FTA® cards for downstream PCR analysis. The results obtained in this tri–national study indicate that solid phase FTA® media can play a role in sample collection, archiving, and transport of samples containing or suspected of containing biological agents, but that FTA® media should be used in support of, and in combination with, liquid phase sampling. Solid phase sample collection using FTA® media and liquid phase sample collection in combination provides several advantages over using either technique alone.
